Induksi Tunas Dari Eksplan Epikotil Jeruk Kasturi (Citrus Microcarpa Bunge.) Dengan Penambahan Bap Dan Kinetin Secara In Vitro
Kasturi orange (Citrus microcarpa Bunge.) is a type of fruit that belongs to the genus citrus, has vitamin C content, and has high antioxidant properties. Generative development after 5 years old. Therefore, by inducing the need for seedlings in large quantities can be achieved in a short time. This study aims to determine the effect of the use of BAP and Kinetin singly or in combination on the formation of shoots from castor epicotyl explants in vitro on MS media and determine the best concentrations of BAP and Kinetin singly or in combination for the formation of shoots from epicotyl explants of castor citrus in vitro on MS media. This study used a completely randomized design (CRD) factorial consisting of BAP (0 mg / l, 0.5 mg / l, 1 mg / l, and 1.5 mg / l) and Kinetin (0 mg / l, 0.5 mg / l, 1 mg / l, and 1.5 mg / l) with 16 combinations and each was repeated 3 (three) replications. The results of this study indicated that the administration of BAP and Kinetin singly or in combination had a significant effect on shoot length, but did not significantly affect the percentage of shoot formation, time of shoot emergence, number of shoots and number of leaves from the epicotyl explants of castor lime. The highest shoot length was at a concentration of 1.5 mg / l BAP + 0.5 mg / l Kinetin at 0.6 cm. While the lowest shoot length was at a concentration of 1.5 mg / l BAP + 1.5 mg / l Kinetin, namely 0.0 cm.
