Seleksi Bakteri Penambat Nitrogen Non-Simbiotik Dalam Mereduksi Logam Fe Dan Identifikasi Menggunakan Sekuen Gen 16s Rrna
Iron (Fe) is one of the essential nutrients needed by plants and microorganisms for their metabolism process. The high level concentration of Fe in the environment can cause toxic effects for living things. Free-living nitrogen fixing bacteria also has tolerance ability to a high level of metal level except for their role as nitrogen fixing. Therefore, this research has to do to selected the ability test of growth these bacteria in Nfb (Nitrogen Free Bromothymolblue) medium with various iron concentration (200, 300, 400, 500, 700, 900, 1000 ppm) and to know their potential for reducing iron and reducing iron test by AAS (Atomic Absorption Spectrophotometer). The potential bacteria were followed by the identification 16S rRNA gene with a universal primer. This research using nitrogen fixing bacteria are NR 10, NR 12, NR 15, NR 18, NR 19, NR 22, NR 24, NR 27, NR 28, NR 30, NR 31, R 41 and R 47. The result showed that only nine bacteria that can grow in Nfb agar+Fe 1000 ppm concentration are NR 15, NR 18, NR 19, NR 22, NR 24, NR 27, NR 28, NR 31, R 47. Three bacterial grow well NR 27, NR 28 and NR 31 have iron reducing power that NR 27 amount to 92,66%, NR 28 amount to 88.29% and NR 31 amount to 81.55%. The identification 16S rRNA gene sequence NR 27 has a similarity number of 99,71% with Ochrobactrum ciceri strain Ca-34.
