Optimasi Waktu Produksi Metabolit Sekunder Berpotensi Antibiotik Dari Media Pertumbuhan Penicillium Sp. Lbkurcc34 Dengan Elisitor Staphylococcus Aureus
Penicillium sp. LBKURCC34 is one of a fungal collection at the Enzyme, Fermentation and Biomolecular Research Laboratory that produce potential secondary metabolites for many applications. Out of same factors affecting the production of secondary metabolites from microorganisms. Incubation time is the most important factor as it gives a considerable influence on the mass of the extract and its biological activity. In this study, secondary metabolite extracts from Penicillium sp. LBKURCC34 were harvested from various incubation time by batch fermentation method in liquid media which Staphylococcus aureus was added as a biotic elicitor on the 3rd day. Identification of secondary metabolite extracts was carried out through the evaporation method to determine the weight of the extract and LC-MS analysis to determine the characteristics of the compound content and antimicrobial activity of the extracts through disc diffusion tests on Escherichia coli, Staphylococcus aureus, Staphylococcus epidermidis, Bacillus substilis, Vibrio parahaemolyticus, Vibrio alginolyticus and Candida albicans. The extracts of secondary metabolites were harvested from 8 days (P8H), 11 days (P11H) and 14 days (P14H) of incubation times and their mass was 110.8 mg, 114.6 mg, and 71 mg, respectively. LC-MS / MS result showed that the peak of the dominant compound in the extracts were generally in the range from 9 to 13 minutes of retention times (from 25 minutes). The data indicated that the characteristics of compounds were more polar. Interestingly, P14H extract contained several less polar compounds at 22.57; 24.32; and 28.17 minutes. The P14H extract contained compound that have higher molecular weight compounds (527-575 Da) with highest activity against E. coli (80.27%). These results indicated that the incubation time is crucial in producing microbial secondary metabolites.
