Analisis Sekuens Dna Penyandi Gliseraldehid-3- Fosfat Dehidrogenase (Gapdh) Pada Tumbuhan Cocor Bebek (Kalanchoe X Laetivirens)
ABSTRACT
Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is a group of
housekeeping genes and its expression in various abiotic stresses is constitutive
and stable. Molecularly, gene expression analysis techniques require internal
controls for basic cell regulation. This study aims to analyze the DNA sequences
encoding GAPDH in cocor bebek (Kalanchoe x laetivirens). The research
procedures included plant morphological observation, sample preparation, total
DNA isolation using Genomic DNA Mini Kit Plant (Geneaid), GAPDH gene
amplification using Polymerase Chain Reaction (PCR) technique, electrophoresis
at 1% agarose and DNA sequence data analysis. The K. x laetivirens GAPDH
DNA sequence obtained in this study is 1107 bp in size and had a similarity of
about 68.96% -73.81% with the GAPDH DNA sequences in several plant species.
The amino acid sequence of K. x laetivirens GAPDH had 5 exons and 5 introns
and had 5 conserved regions. This sequence is the first to be reported from the
species Kalanchoe x laetivirens and genus Kalanchoe. This sequence can be used
for analysis of gene expression in cocor bebek and can be a reference for isolating
the GAPDH gene for other plant species in the genus Kalanchoe and in the
Crassulaceae family.
