Kemampuan Imobilisasi Bacillus Sp. Menggunakan Beberapa Variasi Carrier Untuk Mendegradasi Minyak Bumi
ABSTRACT
Petroleum production activities can cause environmental pollution which has a significant impact on humans and the ecosystem. One way of handling polluted environmental conditions is biodegradation using the Bacillus sp., Which can utilize hydrocarbon compounds in petroleum as a source of carbon to carry out its metabolic activities. Therefore, a study aimed to determine the ability of Bacillus Sp 48 and Bacillus Sp 83 isolates to degrade petroleum by immobilization using oil palm empty bunches (TKKS), sawdust (SG) and rice husks (SP). This research began with the rejuvenation of the bacterial isolate Bacillus sp., with the strain Sp 48 and Sp 83. Test the growth ability of bacterial isolates on Minimal Salt Medium (MSM) + 1% crude oil and pH measurements for a period of 0, 5, 10 and 15 days. Biodegradation test of petroleum with immobilization of bacteria and non-immobilization, measurement of oil content with the Total Petroleum Hydrocarbon (TPH) technique using the gravimetric method and bacterial immobilization test. The results showed that the highest total population of Bacillus Sp 48 bacteria on the 10th day of TKKS treatment was 2,67x109 cfu/ml and SG 2,58x109 cfu/ml. The highest total population of Bacillus Sp 83 bacteria on the 5th day of treatment K was 2,72x109 cfu/ml, TKKS is 2,24x109 cfu/ml, SG is 1,60x109 cfu/ml and SP is 2,68x109 cfu/ml. Bacillus Sp 48 treatment P3.Sp48 had the highest percentage of degradation of 87,95%. Bacillus Sp 83 treatment P3.Sp83 had the highest degradation percentage of 82,47%. Sawdust carriers are the best at accelerating petroleum degradation. The total population of bacteria in the carrier was higher than that in the MSM medium. The highest total bacterial population was on Bacillus Sp 48 by the SG carrier 2,38x1013 cfu/ml and the Bacillus Sp 83 SP carrier 2,54x1013 cfu/ml.
