Deteksi Kandidat Bacterial Key Species Pada Hutan Sekunder Dan Lahan Yang Dialih Fungsi Di Areal Cagar Biosfer Giam Siak Kecil Bukit Batu
ABSTRACT
Biosphere Reserve GSK-BB is an area of peat ecosystem that has been largely converted, causing a decrease in the quality of the peat. Generally, parameters of peat quality use biological, physical and chemical factors. This parameter has not high sensitivity. Therefore, Bacterial Key Species (BKS) was developed from 16S rRNA gene sequences as sensitive parameters in monitoring peat quality. This study aims to find BKS detected from secondary forest and land conversion, specific primer design and in silico primer testing. The method of determination BKS candidates was carried out by selecting sequences based on phylogenetic trees and BLASTn. Furthermore, the primer design and specificity test were carried out using the PCR in silico. The results of the study detected 72 candidates BKS from secondary forest and HTI (BKS-SHI), 93 candidates BKS from secondary forest and rubber plantations (BKS-SKK), and 69 candidates BKS from secondary forest and restoration (BKS-SRE). After analizing with BLASTn, it was found that there were 5 group of candidates BKS-SHI, 9 group of candidates BKS-SKK, 13 group of candidates BKS-SRE. The final result of DNA sequence alignment detected 6 BKS specific candidates (BKS-SHI3, BKS-SKK2, BKS-SRE2, BKS-SRE9, BKS-SRE10, dan BKS-SRE11) which were specific only in secondary forest locations and were not found in land conversion with sequence lenghths of 435-473 bp. The specific primers obtained from the sequences candidates BKS BKS-SHI3 were 6 primers and sequence of candidates BKS-SKK2 was 5 specific primer, while the sequence of candidates BKS-SRE was 11 specific primer. DNA sequence candidates BKS detected has not been found by species name. For this reason, the sequencing of the whole genome to find out names of species from candidates BKS.
