Isolasi Dan Seleksi Antibakteri Aktinomisetes Dari Tanah Hutan Imbo Putui, Desa Petapahan, Kabupaten Kampar
The area “Hutan Adat Imbo Putui” Kampar Regency, Riau is a tropical rain forest
located in Petapahan Village. The soil around the forest area is one of the sources
of actinomycetes diversity. Actinomycetes are microbes with potential in the
medical and pharmaceutical industries because they can produce various secondary
metabolites, one of which is antibacterial. This study aims to isolate and screen
actinomycetes from the soil of the “Hutan Adat Imbo Putui” against bacteria
pathogen. Isolation of actinomycetes using Starch Casein Agar (SCA) medium with
the spread plate method, 3 – 5 days period of incubation. The isolates were further
characterized by macroscopic and biochemical test by Gram staining.
Actinomycetes were selected for antibacterial against Escherichia coli and
Staphylococcus aureus using the Agar Disk method by measuring the zone of
inhibition. The results obtained 27 actinomycetes isolates. Nine isolates had
antibacterial activity against Escherichia coli with the largest inhibition zone
diameter of 26.49 mm (I1L6). Ten actinomycetes isolates were able to inhibit
Staphylococcus aureus with the largest zone of inhibition, namely 28.52 mm
(I6L14), and nine actinomycetes isolates were able to inhibit Escherichia coli and
Staphylococcus aureus bacteria. These isolates have the potential to be developed
to obtain new antibiotics.
