Analisis Profil Pita Rapd Dan Sekuens Carf 005 Pada Kopi Liberika (Coffea Liberica W. Bull Ex Hiern) Resisten Penyakit Karat Daun
Riau Province has liberica coffee varieties which resistant to leaf rust that
has been released by Balittri, namely Liberoid Meranti 1 (Lim 1) and Liberoid
Meranti 2 (Lim 2). Detection of resistance to disease in plants is important to ensure
the identity of varieties and to assist plant breeding programs. This identification
process can be done through a PCR-based molecular approach using random
primers such as RAPD or specific primers. This study aimed to analyze the profile
of the RAPD band and the CARF 005 sequence on Lim 1, Lim 2 and local coffee
from Bengkalis. The RAPD primers used were OPA-04, OPE-03, and OPE-18. The
CARF 005 primer has been reported to amplify the NB-ARC domain in Arabica
coffee. Results of RAPD analysis showed that OPE-03 and OPE-18 generated three
different band patterns that distinguished those three coffee samples. In addition,
OPE-03 produced one band-sized 650 bp specific to Lim 1 and OPA-04 produced
four specific bands in Bengkalis coffee. We suggested that the RAPD marker can
be applied to identity liberica varieties. PCR with CARF 005 marker amplified
fragment containing NB-ARC domain and P-loop/walker A motif in all coffee
samples. These results indicate that the CARF 005 marker can be used to detect leaf
rust resistance in liberica coffee plants.
