Isolasi Dan Uji Penghambatan Α-Glukosidase Metabolit Sekunder Dari Ekstrak N-Heksana Tumbuhan Peperomia Pellucida
Peperomia pellucida is one of species belongs to Piperaceae family which is
widely distributed in Indonesia. This plant commonly is used as a traditional
medicine by various tribes in Indonesia. Several species of this genus are reported
have antidiabetic, antioxidant, antibacterial, and antiinflammatory. This
investigation was intended to isolate P. pellucida secondary metabolites from
n-hexane extract and determine antidiabetic activity of extract and isolated
compound. The plant was extracted by maceration using in methanol. The extract
was then partioned with n-hexane and ethyl acetate. Furthermore, the
n-hexane extract was purified by using vacuum liquid chromatography (VLC)
produced 11 fractions. Fraction 3 and 8 was recrystallized to obtain white needle
crystal with melting point of 142-1440C and white powder crystal with melting
point 252-2540C. The chemical structure was characterized by FTIR spectroscopy
and the results PP-H-03 obtained are considered as β-sitosterol and PP-H-08
considered as daucosterol after spectroscopic data compared to data in the
scientific literature. The extract and the pure compounds were tested for
antidiabetic activity. The results with inhibition persentage values 0,832 % for nhexane
extract, -0,3557 % for PP-H-03 and -1,10281% for PP-H-08. Based from
the data the extract and isolated compound showing weak antidiabetic activity
compared to positive control of acarbose.
