Isolasi Senyawa Metabolit Sekunder Dan Penghambatan Α-Glukosidase Dari Ekstrak N-Heksana Tanaman Miana Hijau (Coleus Hybridus)
Miana (C. hybridus) is an ornamental plant that is widely used as a medicinal
plant. This study aimed to isolate secondary metabolites and test α-glucosidase
inhibition from these plants. Isolation was carried out by maceration method with
methanol and then partitioned with n-hexane solvent to obtain a thick n-hexane
extract. Extracts were separated by vacuum liquid chromatography (VLC) and
purified by flash chromatography. Two isolated pure compounds were obtained
codenamed CHG-H7 and CHG-H8 with melting points of 134º-136ºC and
136º-138ºC, characterized using FTIR spectroscopy. α-Glucosidase inhibitory test
was carried out using the α-glucosidase enzyme and acarbose as positive controls.
The %inhibition screening carried out showed that the %inhibition of n-hexane,
CHG-H7 and CHG-H8 extracts were 49.93%, -1.9889% and 21.267%,
respectively. These results indicate that, the n-hexane extract has potential so that
further IC50 tests were carried out. The results, it is known that the n-hexane
extract of C. hybridus has an IC50 value of 452.62 μg/mL with acarbose having an
IC50 value of 68.410 μg/mL. The comparison of the IC50 value which is quite
high between the extract and acarbose can occur because in the tests carried out,
the acarbose used is a pure compound. Based on the results of the research that
has been done, it can be concluded that the n-hexane extract of C. hybridus has the
potential as an enzyme inhibitor α-glucosidase.
