Induksi Kalus Daun Binahong Merah (Basella Rubra L.) Pada Media Ms Dengan Pemberian 2,4-D Dan Kinetin
ABSTRACT
Red binahong (Basella rubra L.) is a plant that contains medicinal
secondary metabolites. The problem in the development of medicinal plants is
low levels of secondary metabolite. Tissue culture is one solution in producing
secondary metabolites in large quantities. The aims of this research were to
determine the effect and the best combination of 2,4-D and kinetin in inducing
callus on red binahong leaves. The research was condected at the Laboratory of
Plant Biotechnology, Faculty of Agriculture, Univeritas Riau from November
2019 to March 2020 was used a randomized block design with two factors,
namely four levels of 2,4-D (0, 0,5, 1 and 2 ppm) and four levels of kinetin
(0, 0,5, 1 and 2 ppm) with three replications. The results suggested some
significant effect of the interaction of 2,4-D and kinetin on the parameters when
callus appeared and the percentage success of callus formation. The factor of
2,4-D had a significant effect on all parameters. The factor of kinetin had a
significant effect on the parameters when callus appeared and the percentage
success of callus formation. The combination of 0 ppm 2,4-D and 0,5 ppm kinetin
and 1 ppm 2,4-D and 2 ppm had the fastest callus appreance (11,67 HST and
15,17 HST, respectively) and combination of 1 ppm 2,4-D and 2 ppm kinetin had
tend higher callus weight (6,4 mg) and had tend highest percentage success of
callus formation (62,50%). Factor of 2,4-D 2 ppm had the fastest callus
appearance (11,40 HST) and 0,5 ppm had higher callus weight (4,3 mg) and the
highest percentage success of callus formation (50%). Factor of kinetin 0,5 ppm
had the fastest callus appreance (11,42 HST), 2 ppm had tend higher callus weight
(3,7 mg) and 0 ppm had the highest percentage success of callus formation
(37,50%).
