Isolasi Dan Karakterisasi Aktinomisetes Dari Rizosfir Dan Endofit Tumbuhan Siak-Siak (Dianella Ensifolia (L.) Dc.)
Actinomycetes are gram-positive bacteria capable of producing important
metabolite compounds. Isolation uses samples of rizospher and endophyte from the
medicinal plant Dianella ensifolia. The purpose of the study was to obtain isolates
of actinomycetes from rizosfir and endophyte D. ensifolia and their
characterization. Isolation uses three pre-treatment methods in rizosfir samples and
uses 3 parts in the form of roots, rhizomes and leaves from the D. ensifolia plant to
isolate endophyte activityinomycetes. Characterization includes macroscopic,
microscopic, and biochemical tests. The results of the study obtained 16 isolates
from actinomycetes rizosfir, 9 isolates obtained from pre-treatment of wet heat and
without pre-treatment as many as 7 isolates. Endophyte insulation results are not
obtained isolated actinonomisetes. Macroscopic characterization of insulated
actinomycetes consists of small and moderate size, irregular, and circular shape,
entire colony edge, serrate, and undulate, elevation of umbonate, raised, flat, and
convex. The characterization of microscopic actinomycetes in the form of whole
isolates is gram-positive bacteria and has a chain of polyspores. A total of 14
isolates were able to produce amylase enzymes, 2 isolates were able to produce
gelatinase enzymes, and 9 isolates were able to produce catalase enzymes.
