Penambahan Air Rebusan Kentang Pada Media Murashige And Skoog (Ms) Terhadap Pertumbuhan Protokorm Dari Biji Anggrek Sendu (Grammatophyllum Stapeliiflorum) Secara In Vitro
The Sendu Orchid (Grammatophyllum stapeliiflorum) is one of the most popular
orchid species because of its beauty. The sendu orchid is characterized by the
shape of the flower stalk that sticks down and the brown flowers with white and
yellow spots that look like drops of water or sad. This type of orchid is almost
extinct and is rarely found in its natural habitat, so it is necessary to propagate
orchids in vitro. The seed culture method is one of the most effective methods for
the propagation of rare orchids. This study aims to determine the effect of adding
potato boiled water on protocorm growth and determine the best concentration for
the growth of G. stapeliiflorum protocorm in vitro. This study used a Completely
Randomized Design (CRD) with the addition of potato boiled water
(concentrations 0, 50, 100, 150 and 200 ml/L) on Murashige and Skoog (MS)
media. The results showed that the percentage of live explants was 100% in all
treatments, indicated by the change of white to greenish seeds to form glossy and
dense globular (Phase 1). The best concentration of potato boiled water for the
growth of the protocorm of G.stapeliiflorum orchids in vitro on MS media was at
a concentration of 150 ml/L which indicated the highest increase in PLBs growth
compared to other potato boiled water treatments and controls with an average of
60.60 DAP.
