Pengaruh Bap, Naa Serta Kombinasi Bap Dan Naa Terhadap Perkembangan Kultur Meristem Apikal Sagu Duri In-Vitro
ABSTRACT
Sago is a food crop that has many advantages over other food crops. There are three
types of sago found in Meranti Islands Regency, namely Duri sago, Sangka sago,
and Bemban sago. The type of sago Duri is the most widely distributed and has a high
starch production. The propagation of sago plants is constrained by the availability
of uniform sago seeds in large quantities. One of the ways to overcome this problem is
by propagation of tissue culture through apical meristem culture of sago Duri with the
addition of cytokinins and auxins on MS media. The purpose of this study was to
determine the effect of BAP, NAA and the combination of BAP and NAA on MS media
and to obtain the best treatment for shoot morphogenesis of shoot of sago thorn apical
meristem explants in vitro. The study used a completely randomized design consisting
of 8 treatments and 3 replications so that there were 24 experimental units.
The parameters observed were when the shoots appeared, the number of shoots, shoot
height and the percentage of growth success. Data analysis used analysis of variance
and further tested using DNMRT level 5%. The results showed that when shoots
appeared, the apical meristem explants of sago Duri tended to be faster in the treatment
of BAP 2 mg/l + NAA 2 mg/l, which was 60.17 DAP. The number of shoots tended
to be more in the 1 mg/l BAP treatment, namely 1.83 shoots. Shoot height tended to be
higher in the treatment of BAP 1 mg/l, BAP 2 mg/l and NAA 2 mg/l, namely 11.83 mm.
The percentage of success in growing with the administration of NAA 1 mg/l and
BAP 1 mg/l + NAA 1 mg/l tends to be higher, namely 66.67%.
