Analisis Sekuens Barkode Dna Pada Daerah Matk Tumbuhan Kabau (Archidendron Microcarpum)
Kabau in the Riau area is not widely cultivated and only grows wild in
primary forest and secondary lowland forests in Indonesia. This condition makes
kabau plants can be endangared. The development of technology to identify plant
species can be performed molecularly using short pieces of DNA called DNA
barcodes. Therefore, an exploration, such as building a database of DNA sequence
from kabau plants is necessary. Analysis od DNA barcodes sequence in the matK
area in kabau plants (Archidendron microcarpum) will be conducted in this study.
The stages of this study include sampling, total DNA isolation, Polymerase Chain
Reaction, electrophoresis, sequencing and data analysis using BioEdit version 7.0,
BLASTn and MEGA version 6.0. The results obtained from this study are the
length of the matK sequence in kabau plants is 866 bp and has the highest
similarity value with Pithecellobium clypearia, which is 99.31%. This study
showed that there were 22 nucleotide variations and 4 of them were critical
nucleotides. None of the accessions had a 100% similarity value, this indicates
that the DNA sequence of the matK gene from kabau plants is not yet available in
the database GenBank.
