Analisis Sekuens Barkode Dna Pada Daerah Gen Matk Tumbuhan Petai (Parkia Speciosa)
DNA barcoding is a technique that uses standard short pieces of DNA or
also known as DNA barcodes whose ability has been tested to distinguish at the
species level. One of the DNA barcodes in plants is the DNA barcode on the matK
gene. The purpose of this study was to analyze the DNA barcode sequences in the
matK gene area of petai (Parkia speciosa) taken from Siak Hulu District, Kampar
Regency, Riau Province. The research stages consisted of morphological
observations, sample preparation, total DNA isolation of young and fresh petai
leaves, total DNA electrophoresis, Polymerase Chain Reaction (PCR),
electrophoresis of PCR products, sequencing and data analysis. Data analysis
using Bioedit, BLASTn and MEGA6 programs. The results showed that the DNA
sequence of the matK gene of the petai plant was 836 bp in size and had a 100%
similarity or percent identity with Parkia speciosa and Parkia biglandulosa.
There are 18 nucleotide variations in the DNA sequences of the petai plant sample
with the DNA sequences of ten other species. Based on the dendogram, petai
plants form a group with Parkia speciosa, Parkia biglandulosa, Parkia multijuga,
and Parkia timoriana.
