Isolasi Dan Karakterisasi Bakteri Xanthomonas Sp. Penyebab Penyakit Hawar Daun Bakteri Pada Tanaman Bawang Merah (Alliium Ascalonicum L.)
ABSTRACT
Shallot (Allium ascalonicum L.) is a horticultural plant that contains
carbohydrates, fats, vitamins and minerals. The productivity of shallots are low in
Riau Province, which is caused by Xanthomonas sp. This bacterium is the
pathogen of the important disease that attack at various growth stages in shallot
plants. The Symptoms is yellowish white spots, chlorosis to shoot death. The
losses caused by these bacteria are very large, so proper control is needed. This
research aims to isolate and characterize Xanthomonas sp. cause of bacterial leaf
blight on shallot plants. Data on morphological characteristics, hypersensitivity
reactions, Koch's postulates and biochemical characteristics were analyzed
descriptively and presented in the form of figures and tables. The results of the
isolation of bacteria that have macroscopic similarities were obtained as many as
10 isolates (X1, X2, X3, X4, X5, X6, X7, X8, X9 and X10). Isolates X2 and X3
showed symptoms of necrosis in tobacco plants, symptomatic of chlorosis on
Koch's postulate test, negatively reacted on gram and oxidase tests and positively
reacted on catalase and levan formation tests.
