Analisis Sekuens Barkode Dna Daerah Gen Matk Tumbuhan Jengkol Papan (Archidendron Pauciflorum)
Jengkol papan (Archidendrom pauciflorum) is a plant member of Fabaceae family
which contains various nutrients and chemical compounds, and has a fruit with a
unique smell. DNA barcodes are short and unique DNA sequences of a gene that
are used as a fast and precise alternative in identifying organisms. The matK gene
is a gene that has a high rate of nucleotide substitution and a variety of sequence
sequences. This study aims to analyze the DNA barcode sequences of the matK
gene of the jengkol papan plant (Archidendron pauciflorum). The research method
started from taking samples of jengkol papan leaves and then observing
morphological characters for making descriptions. Furthermore, molecular analysis
starting from the sample preparation stage, total DNA isolation, total DNA
electrophoresis, PCR, electrophoresis of PCR products, sequencing, and data
analysis using BioEdit software version 7, BLASTn and MEGA 6.0. The matK gene
sequence of jengkol papan plant obtained has a size of 839 bp. And has the closest
kinship with Archidendron lucyi with an identity value of 99.28%. The level of
nucleotide variation of the jengkol papan sample with other accessions was 23
nucleotide variations and five of them were critical nucleotides. Critical nucleotides
are located at nucleotide numbers 367, 716, 821, 828 and 836.
