Analisis Sekuens Barkode Dna Pada Daerah Trnl-Trnl-Trnf Intergenic Spacer Tumbuhan Petai (Parkia Speciosa)
DNA barcoding is a method used to identify species using short DNA
sequences from the genome of an organism. One of the DNA barcodes commonly
used in molecular analysis is the trnL-trnL-trnF Intergenic Spacer, because it has
a high mutation rate and variability. This study aims to analyze the barcode
sequence trnL-trnL-trnF Intergenic Spacer in petai (Parkia speciosa) taken in
Siak Hulu District, Kampar Regency, Riau Province. The methods in this study
consisted of isolation of total DNA from young and fresh leaves of petai plants,
total DNA electrophoresis, PCR, electrophoresis of PCR products, sequencing and
data analysis using software such as Bioedit, BLASTn, and MEGA. The results of
this study showed that the DNA sequence of trnL-trnL-trnF Intergenic Spacer in
petai plants obtained a size of 969 bp and had the highest similarity with Parkia
biglandulosa. This is also supported by a high maximum score (1746), low EValue
(0.0), high percent identity (100%) and high query cover (99%). The results
of the study showed that there were 71 nucleotide variations among the accessions
studied. Petai plants form a group with Parkia biglandulosa, Parkia speciosa,
Parkia ulei, Stryphnodendron duckeanum, Piptadenia floribunda and Piptadenia
minutiflora.
