Longan (Dimorcapus longan) is a fruit-producing plant that can grow in subtropical and tropical areas, one of which is in Indonesia. Mata kucing longan (Dimocarpus longan subsp. malesianus Leenh) is a subspecies of longan. Mata kucing longan is one of the riches of flora that is not found in every region. This plant is native to Indonesia which is spread across various regions such as Kalimantan, Java and Sumatra. Analysis of longan diversity at the morphological and molecular levels is expected to be able to explain the general kinship relationship, seen from the genetic distance between the two. Morphological characterization is often influenced by environmental factors, so the use of molecular markers is expected to provide a description of the characterization with a fairly high accuracy in seeing the kinship relationship. Molecular analysis of plant kinship is very useful in plant breeding programs such as in selecting parents or improving varieties. This study aims to determine the closeness of the kinship relationship between longan and cat's eye longan based on RAPD markers.
This research was conducted from January 2023 to March 2023 at the Laboratory of Genetics and Molecular Biology, Faculty of Mathematics and Natural Sciences, University of Riau. Longan samples (sample code K) and mata kucing longan (sample code MK) and each represented by nine individuals were taken from Peranap District, Indragiri Hulu Regency, Riau Province. This study consists of several stages, namely sample collection, total DNA isolation, electrophoresis, PCR, and data analysis. Data were analyzed using the UPGMA (Unweighted Pair Group Method with Arithmetic Average) method using MVSP 3.22 software. DNA isolation was performed using the Genomic DNA Mini Kit Plant (Geneaid). The primers used in this study included OPD-08, D-08, X-01, and OPJ-20.
The results showed a total of 40 RAPD bands obtained in this study. The OPD-08 primer produced the longest band, which was 2500 bp. The highest similarity coefficient reached 77.78%, which was obtained between the mata kucing genotypes of samples 3 (MK3) and 4 (MK4). The lowest similarity value was found in several samples, namely between longan sample 2 (K2) and cat's eye sample 9 (MK9), longan sample 3 (K3) and mata kucing sample 1 (MK1), longan sample 3 (K3) and mata kucing sample 2 (MK2), longan sample 3 (K3) and mata kucing sample 6 (MK6) of 0.000%. The percentage of kinship between longan plants and mata kucing longan is 36%. Both are genetically different, meaning they do not have a close kinship.
