Analisis Gen Cytb dan D-Loop Region Pada Ikan Baung Pisang (Bagroides melapterus, Bleeker, 1851) Asal Sungai Kampar Kecamatan Langgam, Provinsi Riau)
Baung pisang fish (Bagroides melapterus) is one of the fish with high eco-nomic value that found in Kampar River, Langgam District, Riau Province. The availability of genetic information baung pisang fish is still limited, but the popula-tion of baung pisang has decreased, especially baung pisang fish (B. melapterus) found in the Kampar River, Langgam District, Riau Province. This study aims to analyze the cytb gene sequence and the D-Loop region that can be used as a genetic marker of baung pisang fish (B. melapterus) originating from the Kampar River, Langgam District, Riau Province. The methods included sampling of fish, DNA extraction from the muscles of baung pisang fish using Tissue Genomic Mini Kit (Geneaid), Polymerase Chain Reaction (PCR), electrophoresis, and sequenc-ing. The obtained data were analyzed using MEGA software version 11. Sequence analysis result of the cyt b gene of baung pisang fish with a length of 651 bp. The three of baung pisang fish samples had the highest similarity with Hemibagrus wyckiodes (91.04%). There are 59 nucleotide variations among the accessions studied and 20 of them are identical nucleotides and there are 20 amino acid variations and 5 of them are identical amino acid. These variation are caused by changes of nucleotide 1 and 2 of bases. The results of D-loop region sequence with a length of 802 bp. The three baung pisang fish samples have the highest similarity with Leiocassis longirostris (80.86%). There were 302 nucleotide variations among the accessions studied and 72 of them were identical nucleotides.
