Isolasi Senyawa Metabolit Sekunder Ekstrak N-Heksana Dan Uji Antijamur Dari Tumbuhan Paku (Drynaria Sparsisora)
Drynaria sparsisora, commonly known in Java as “simbar layangan” or “paku langlayang”, belongs to the family Polypodiaceae. This plant is characterized by an upright rhizome with a tapering base and dense scales, as well as dimorphic leaves. It is often found growing on rocks, in open areas such as riverbanks, and on tall trees. This study aimed to isolate secondary metabolites from the n-hexane extract of D. sparsisora and to evaluate their antifungal activity using the microdilution method against Candida albicans, Candida krusei, and Candida glabrata. The viscous n-hexane extract was fractionated using Vacuum Liquid Chromatography (VLC), resulting in five major fractions that were not yet pure. Fraction 1, the primary VLC fraction, was further separated using flash column chromatography, yielding 29 subfractions. Purity analysis of subfraction 25 using thin-layer chromatography (TLC) revealed a single spot under UV light at wavelengths of 254 nm and 365 nm, with positive staining using anisaldehyde reagent. The purity of this subfraction was further supported by its melting point range of 80–82°C and the presence of a single dominant peak in the High-Performance Liquid Chromatography (HPLC) chromatogram. This purified subfraction was designated as compound DS-HX-F1-SF25. Structural characterization was conducted using UV-Vis and FTIR spectroscopy, which indicated that DS-HX-F1-SF25 belongs to the phenolic compound group. The n-hexane extract, its fractions, and the purified compound were tested for antifungal activity using the microdilution method against three pathogenic fungi: C. albicans, C. krusei, and C. glabrata. The antifungal assay results showed that the n-hexane extract exhibited a Minimum Inhibitory Concentration (MIC) of 1000 ppm against C. krusei and C. glabrata. Fractions 1–5 demonstrated an MIC of 250 ppm against C. glabrata, while the pure compound DS-HX-F1-SF25 showed an MIC of 100 ppm against both C. krusei and
C. glabrata. The Minimum Fungicidal Concentration (MFC) of the n-hexane extract was 2000 ppm against C. albicans. Fractions 1–5 exhibited fungicidal activity at concentrations of 500–1000 ppm against C. krusei. However, DS-HX- F1-SF25 did not exhibit any fungicidal activity (MFC) at the tested concentrations.
