Optimasi Biosorpsi Methylene Blue Dengan Immobilized Microalgae Beads (Chlorella Sp.) Menggunakan Response Surface Methodology

ABSTRACT
Methylene blue synthetic dye is carcinogenic, toxic, and flammable so it needs to
be processed before being discharged into the environment. In this study, methylene
blue was processed by biosorption method using immobilized Chlorella sp.
microalgae (Immobilized Microalgae Beads / IMB). This study aims to obtain the
optimum variables and empirical model of IMB in the removal of methylene blue
dye and optical density in liquid waste using Response Surface Methodology (RSM)
with Box Behnken Design (BBD) technique and obtain the physiochemistry of IMB
before and after treatment. RSM with BBD technique is used to obtain the optimum
conditions in the response. The research factors used include methylene blue
concentration (15-25 mg/L), adsorbent dose (6-10 g/L), and contact time (1-5
hours). The optimum conditions for removal efficiency and optical density of
methylene blue were obtained at a concentration of 15 mg/L, an adsorbent dose of
10 g/L, and a contact time of 5 hours with a removal efficiency of 55.6% and an
optical density of 2.620. The SEM-EDS test results showed that the IMB after
biosorption had a rough, uneven surface, and cells tended to be flat and there was
an increase in Cl and S elements because the active area of the beads was filled
with methylene blue molecules due to electrostatic interactions. FTIR identified that
methylene blue tended to interact with the –OH and C=O groups. This interaction
occurred through hydrogen bonds and the involvement of carbonyl groups with
methylene blue molecules. The mechanical strength test produced values before and
after processing with values of 60.56 gForce and 81.87 gForce. This increase
occurred because the ionic interaction of methylene blue with functional groups
caused matrix strengthening so that the beads were denser and more compact.
