Pengaruh Kombinasi Bap Dengan Ekstrak Tomat Terhadap Pertumbuhan Eksplan Epikotil Jeruk Siam (Citrus Nobilis Lour.) Asal Kampar Secara In Vitro
ABSTRACT
This study aims to determine the effect of the best concentration of the
combination of BAP with tomato extract on the growth of epicotyl explants of
Citrus nobilis Lour. and to support the growth of Citrus nobilis Lour. plantlets
from Kampar in vitro. This research was conducted at the Biotechnology
Laboratory, Faculty of Agriculture, Riau University, Pekanbaru from Desember
2024 to Mei 2025. This research is a non-factorial experiment arranged according
to a complete randomized design (CRD). The treatments in this study were: Z1:
BAP 0.25 ppm + tomato extract 20%, Z2: BAP 0.5 ppm + tomato extract 15%,
Z3: BAP 0.75 ppm + tomato extract 10%, Z4: BAP 1 ppm + tomato extract 5%,
and Z5: BAP 1.25 ppm + tomato extract 0%. The parameters observed were
number of increase in shoot height, leaves increment, root emergence, root length,
and percentage of living explants. Observation data on root emergence, root
length, and percentage of living explants were analyzed descriptively. Observation
data on shoot height increase and leaf number increase were analyzed using
analysis of variance and continued with duncan's new multiple range test
(DNMRT) at a 5% confidence level. Data analysis was carried out using the
statistical package for the social sciences (SPSS) software. The results of the study
showed that the treatment of BAP 1.25 ppm + 0% tomato extract gave the best
response to the increase in shoot height of 1.37 cm and the percentage of living
explants of 93.75%, the treatment of BAP 1 ppm + 5% tomato extract gave the
best response to the increase in the number of leaves of 5.31 strands, but almost
all treatments cannot produce roots, but instead produce callus
