Seleksi Dan Uji Antagonisme Isolat Jamur Rizosfer Tanaman Manggis (Garcinia Mangostana L.) Terhadap Phytophthora Palmivora Penyebab Penyakit Busuk Pangkal Batang Pada Tanaman Jeruk Siam Serta Identifikasinya
ABSTRACT
The aim of this research is to select and obtain isolates of rhizosphere fungi from mangosteen plants which have high antagonism activity against Phytophthora palmivora, the cause of stem base rot disease of siamese citrus plant and to identify to the genus level. The study was conducted at the Laboratory of Plant Disease, Faculty of Agriculture, University of Riau, Pekanbaru. The research methods used in this study were exploration, observation and experimentation which consisted of isolation and purification of rhizosphere fungi from mangosteen plants using the exploration method, hypovirulence test of the isolates using the observation method, growth rate test of the isolates colonies which have a high inhibition ability against P. palmivora using experimental method, hyperparasitism interaction test of the high inhibition ability isolates against P. palmivora isolate using observation method, and the identification of the isolates against using observation method. The results of the research obtained 7 isolates, 6 isolates were hypovirulent and 1 isolate was virulent. Isolate R3 had the highest inhibition ability (89.33%), the biggest colony diameter (36.11 mm) and the fastest growth rate (23.33 mm/day), respectively. There were different types of hyperparsitism interaction of the rhizosphere fungi, namely lysis (isolate R4), entanglement (isolate R3) and attachment (isolate R1). Identification of the rhizosphere fungi isolates showed that isolate R3 belonged to the genus of Trichoderma and isolates R1 and R4 belonged to the genus of Mucor. Those three isolates have high antagonism activities and potential to become a biological agency to control P. palmivora on of siamese citrus plants.
