Selaginella doederleinii Hieron, commonly known as "paku cakar ayam," is a plant from the Selaginellaceae family that known for its anticancer and antioxidant properties, its antibacterial potential has not been extensively investigated. This study aimed to isolate secondary metabolite compounds from the n-hexane extract of S. doederleinii and to evaluate their antibacterial activity using the microdilution method against Staphylococcus aureus, Bacillus cereus, Salmonella typhimurium, and Vibrio parahaemolyticus. Extraction was performed through successive maceration. The n-hexane extract was further fractionated via Vacuum Liquid Chromatography (VLC), resulting in six main fractions. Further separation of fraction 1 using flash column chromatography produced 16 subfractions, with subfraction 9 purified further to yield 7 subfractions. From subfraction 3, a white crystalline compound (12 mg) coded as CA-H-F1-SG9-SF3 was isolated. Its purity was confirmed by thin-layer chromatography (TLC) using a three-solvent system, which showed a single spot under UV light at 254 and 366 nm, as well as under anisaldehyde staining. The melting point of CA-H-F1-SG9- SF3 was recorded at 66–68°C and HPLC analysis revealed a one dominant peak at a retention time of 4.544 minutes, which indicated that the CA-H-F1-SG9-SF3 compound was pure. Characterization using UV-Vis, FTIR, and phytochemical analyses with FeCl₃ suggested that CA-H-F1-SG9-SF3 belongs to the phenolic class. Antibacterial testing revealed that the methanol extract exhibited the strongest activity against S. aureus, with a minimum inhibitory concentration (MIC) of 250 ppm and a minimum bactericidal concentration (MBC) of 500 ppm. The isolated compound CA-H-F1-SG9-SF3 was more effective against Gram- positive bacteria but not better than the positive control (chloramphenicol) with a MIC value of 25 ppm for each bacteria.
