Induksi Kalus Daun Pegagan (Centella Asiatica L. Urban) Pada Media Ms Dengan Pemberian 2,4-D Dan Bap Secara In Vitro
ABSTRACT
Pennywort (Centella asiatica L. Urban) is a plant that contains medicinal secondary metabolites. Tissue culture is one solution in producing secondary metabolites in large quantities. The objectives of this research were to determine the effect and the best combination of 2,4-D and BAP in inducing callus on pennywort leaves. The research was used a randomized block design with two factors, namely 2,4-D with five levels of 0 ppm, 0,5 ppm, 1 ppm, 1,5 ppm, 2 ppm and BAP with five levels of 0 ppm, 0,5 ppm, 1 ppm, 1,5 ppm, 2 ppm. The results suggested all significant effects of giving factor 2,4-D and BAP and the interaction of 2,4-D and BAP on all the parameters. The combination of treatment 1,5 ppm 2,4-D and 2,0 ppm BAP had the fastest callus appearance (4,67 HST). The combination of treatment 0 ppm 2,4-D and 1 ppm BAP had some callus texture which was crumbs and compact and had a lot of callus growth. The combination of treatment 0 ppm 2,4-D and 2 ppm BAP had a high percent of successful formation of callus (40%), high percentage of live explants (40%) and had a lot of callus growth. The combination of treatment 0 ppm 2,4-D and 0,5 ppm BAP had the highest percentage of callus formation success (53,33%) and had some callus texture which was crumbs, compact and combination between crumbs and compact and had the highest percentage of live explants (66,67%)
