Isolasi Dan Identifikasi Senyawa Metabolit Sekunder Ekstrak N-Heksana Dari Daun Tumbuhan Akar Kuning (Arcangelisia Flava (L.) Merr)
Arcangelisia flava, commonly known as akar kuning, is a medicinal plant belonging to the family Menispermaceae, and is well-known especially among the Dayak tribe as a traditional remedy for various diseases. The isolation process began with the extraction of the leaves of A. flava using the maceration method. The n-hexane extract of the plant's leaves was purified using vacuum liquid chromatography (VLC), yielding five main fractions. Fraction 3 from the VLC process was selected for further fractionation using flash chromatography, resulting in 60 subfractions. Among these, subfractions 10 and 50, which appeared as white crystals and yellow amorphous (50 mg and 8 mg respectively), were suspected to be pure and were labeled AK-NT-F3-SF10 and AK-NT-F3-SF50. These two isolates were identified as beta-sitosterol and an indole-type alkaloid, respectively. A combination of subfractions 35–45 from the initial flash chromatography was further fractionated using flash chromatography, resulting in 35 new subfractions. Subfraction 30, which yielded a white solid (20 mg), was labeled AK-NT-SF35-GF30, suspected to be pure and identified as the compound daucosterol. A combination of fractions 5–9 from the initial flash chromatography was further purified using recrystallization, producing two isolates AK-NT-F3-SF2-R1 and AK-NT-F3-SF2- R2, appearing as white crystals weighing 63 mg and 34 mg respectively. AK-NT- F3-SF2-R1 was identified using GC-MS instrumentation as a mixture of α-amyrin and β-amyrin compounds. AK-NT-F3-SF2-R2 was identified as a steroid compound based on a phytochemical test using the Liebermann–Burchard reagent. A combination of subfractions 46–48 from the initial flash chromatography was further purified using liquid-liquid extraction, yielding an isolate in the form of a yellow solid (83 mg), labeled AK-NT-F3-SF45-R. This isolate was identified as an alkaloid compound.
