Respon Eksplan Tangkai Daun Dari Planlet Tacca Chantrieri André Dengan Penambahan 6-Benzyl Amino Purine (Bap) Secara In Vitro
Tacca chantrieri André is an exotic ornamental plant with potential medicinal properties, however it has limitations in generative propagation due to its low germination rate and slow growth, making in vitro propagation necessary. Tissue culture is considered an effective method for shoot multiplication of T. chantrieri. This study aims to determine the effect of BAP application and the optimal BAP concentration on the response of leaf stalk explants from T. chantrieri planlets grown in vitro. This study employed a Completely Randomized Design (CRD) consisting of four treatments: (0 mg/L), 0.5 mg/L, 1 mg/L, and 1.5 mg/L BAP, each replicated seven times. The results demonstrated a 100% explant survival rate across all treatments. BAP supplementation significantly influenced all measured parameters. The optimal BAP concentration was found at the 1 mg/L treatment, which resulted in 100% explant survival, the fastest shoot emergence (9.43 days after planting), the highest number of shoots (11.29 shoots), the earliest leaf emergence (24.29 days after planting), and the greatest number of leaves (4.82 leaves).The application of BAP plays a crucial role in stimulating cell division and initiating shoot and leaf formation in T. chantrieri explants under in vitro conditions.
