ABSTRACT
Koto Panjang Reservoir was built in early 1993 with the main purpose as a
hydroelectric power plant (PLTA). The reservoir area of around 12,400 Ha is
widely used for various other purposes, including flood control, transportation,
tourism and fisheries. The fisheries business that is quite developed in the Koto
Panjang Reservoir is fish farming in floating net cages (KJA). The utilization of
reservoir waters for fish farming activities in KJA has started since 2006 and until
now the number has continued to increase.
In fish farming in KJA, the use of pellets as fish feed is not entirely eaten
by the fish. The feed waste that is thrown into the water is estimated at around 30-
40%. Fish feed that is not consumed by the fish will be thrown into the water
body as waste and accumulate at the bottom of the waters. The accumulation of
organic waste from leftover feed and metabolism will undergo decomposition and
break down into nutrients, especially nitrogen (N) and phosphorus (P)
compounds. High nitrogen and phosphorus compounds in wastewater effluent can
have negative impacts on receiving water bodies such as reducing dissolved
oxygen content, triggering eutrophication and increasing the toxicity levels of a
water body. If this continues for a long period of time, it will have an impact on
the decline in water quality.
Daily community activities have the potential to have a negative impact on
aquatic ecosystems, namely the entry of pollutants and waste from economic
activities such as motorboats. The entry of these pollutants into the waters can
cause a problem, one of which is pollution by the heavy metal lead (Pb).
This study aims to determine the types of bacteria that accumulate heavy
metal Lead (Pb) in carp (Cyprinus carpio) in floating net cages in the Koto
Panjang reservoir which is used as a Pocket Book design in biology learning for
class X SMA. This research is a descriptive study with an experimental method
carried out from October to November 2024 at the Microbiology Laboratory of
the Faculty of Medicine. This research consists of two stages, namely the stage of
collecting data on isolation and characterization of lead (Pb) heavy metal bacteria
in carp and the stage of designing a pocket book. Isolation of lead (Pb) heavy
metal bacteria in carp using Mueller Hinton Broth (MHB) media. Characterization
of lead (Pb) heavy metal bacterial isolates in carp was carried out by macroscopic
(morphological), microscopic (gram staining) and biochemical tests. The data
obtained were then analyzed descriptively and presented in the form of tables and
figures. The results of the study were interpreted into a pocket book in biology
learning for class X SMA on the concept of Environmental Change and
Conservation regarding environmental pollution.
The results of the study can be concluded that the bacterial isolates PbIiWa
1, PbIiWa 2, PbUIWa1, and PbIIWa2 have the same characteristics in shape,
namely circular, entire edge, convex elevation. The bacterial isolate PbIiWa1 has
a different edge shape, namely curled. The bacterial isolates PbIiWa1 and
PbUiWa2 have different colors, PbIiWa1 is clear while the bacterial isolate
PbUiWa2 has a different color, namely greenish.
The results of microscopic observations obtained the results of isolates of
lead (Pb) heavy metal tolerant bacteria with the same cell shape variants. Isolates
PbIiWa1, PbIiWa2, PbUIWa1, and PbUIWa2 are categorized as gram-negative
bacteria because they are red and have a bacillus rod-shaped cell. The results of
the isolation and identification of lead (Pb) heavy metal bacteria in goldfish can be
used as additional learning materials for students, especially in biology learning
for class X SMA on the main material of Environmental Change and
Conservation regarding Environmental Pollution.
