Aktivitas Antidepresan Ekstrak N-Heksana Biji Pinang Tua (Areca Catechu L.) Dengan Metoda Forced Swim Test Pada Mencit Jantan (Mus Musculus) Yang Diuji Secara Simultan
Depression is a mental health disorder that often occurs in society, which is characterized by unstable mood swings. Synthetic antidepressants are quite widely available, these antidepressant drugs have side effects so that alternative antidepressant drugs from herbal plants are needed which have fewer side effects. One of the plants that has the potential as an antidepressant is areca nut. The purpose of this study was to determine the best dose of antidepressant activity from n-hexane extract of old areca nut seeds that were tested simultaneously. In this research, it was started with n-hexane extraction from old areca nut seeds and continued with phytochemical tests. The method used is the Forced Swim Test (FST). FST method consists of acute and sub-chronic FST with 2 test models, namely the FST test in single animals and multiple animals simultaneously. Acute FST was carried out by administering antidepressants to mice, namely n- hexane extract with various doses (10, 50 and 100 mg/kg), fluoxetine 20 mg/kg and saline (NaCl 0.9%). After obtaining the best dose, it was continued with sub- chronic antidepressants for 7 days. On day 7, sub-chronic FST was performed and ended with a toxicity test. The results of the phytochemical test of the n- hexane extract of old areca nut seeds showed positive results from the content of secondary metabolites, namely saponins and steroids. Old areca nut n-hexane extract at a dose of 50 mg/kg has antidepressant activity as can be seen from the results of the percentage reduction in immobility time of single test animals which was significantly different from that of multiple test animals (18.39%; 24.88%). n-hexane extract of old areca nut seeds at a dose of 50 mg/kg given to mice for 7 consecutive days did not show any toxic effects.
