Identifikasi Dan Uji Patogenitas Bakteri Asam Laktat ( BAL ) Dari Saluran Pencernaan Belut Sawah (Monopterus Albus) Yang Dipelihara Pada Sistem Bioflok
This research was carried out from July to October 2021 at the
Laboratory of Fish Parasites and Diseases, Faculty of Fisheries and Marine,
Riau University. The purpose of this study was to identify species of lactic acid
bacteria present in the digestive tract of Asian Swamp Eel (Monopterus albus)
and to test the activity of these lactic acid bacteria against pathogenic bacteria
such as Aeromonas hydrophila, Staphylococcus aureus, Pseudomonas sp. and
Vibrio sp. and testing the pathogenicity of lactic acid bacteria on fish. While, the
benefits of this research can provide a scientific contribution to the development
of science in the field of fish health and aquaculture.
The material used was Asian Swamp Eel (Monopterus albus) obtained
from a farmer in Pekanbaru and isolates of pathogenic bacteria A. hydrophila, S.
aureus, Pseudomonas sp. and Vibrio sp. from the collection of the Laboratory of
Fish Parasites and Diseases, Faculty of Fisheries and Marine, Riau University.
The method used is the experimental method. This research is a follow-up study
on the maintenance of Monopterus albus with a biofloc system. Isolation of Lactic
Acid Bacteria was carried out on the best research results in P3 treatment with a
stocking density of 30 ind/25 liters of water. Isolation of lactic acid bacteria was
carried out from the digestive tract of Asian Swamp Eel (Monopterus albus).
Then, identification of bacteria is carried out by physical, biochemical and
molecular tests to determine the type of lactic acid bacteria isolated. The
identified bacteria were then tested for antimicrobial activity against pathogenic
bacteria like aA. hydrophila, S. aureus, Pseudomonas sp. and Vibrio sp. After
that, the pathogenicity test was carried out on the test fish, carp and catfish.
The results showed, 3 types of lactic acid bacteria identified from the
intestine of Asian Swamp Eel (Monopterus albus) which were reared with a
biofloc system and these bacteria were antagonistic to the pathogenic bacteria A.
hydrophila, S. aureus, Pseudomonas sp. and Vibrio sp. The three isolates had
clear zones against A. hydrophila (12-16 mm), S. aureus (12-14 mm),
Pseudomonas sp. (11-13 mm) and Vibrio sp. (10-12 mm). The results of the
Molecular Test with the PCR Method obtained bacteria, including: Bacillus sp.
with 99% homology, Bacillus subtilis 100% homology, and 99% homology
Paenibacillus cookii. The results of the three isolates were not pathogenicity,
which was indicated by an SR level of 100%, so that, the bacteria identified
(Bacillus sp., Bacillus subtilis and Paenibacillus cookii) could be used as
probiotic candidates.
