Isolasi Senyawa Metabolit Sekunder Ekstrak N-Heksana Dan Uji Antibakteri Dari Tumbuhan Paku (Drynaria Sparsisora)
Drynaria sparsisora, commonly known as the "langlayang fern," is a member of the Polypodiaceae family. This plant typically grows on rocks, in open areas, along riverbanks, and can also exist as an epiphyte on tall trees. Its leaves are characterized by fertile margins with a pinnate venation pattern, and the central part of the petiole is broad, short, and thin. This study aims to isolate secondary metabolites from the n-hexane extract of D. sparsisora and evaluate their antibacterial activity using the microdilution method against Staphylococcus aureus, Bacillus cereus, Salmonella typhimurium, and Vibrio parahaemolyticus. The n-hexane extract was initially fractionated using vacuum liquid chromatography (VLC), resulting in five main fractions, which were all impure. Fraction 1 was further purified using flash chromatography, yielding 19 sub-fractions, which also remained impure. Sub-fraction 15 underwent another purification step via flash chromatography, yielding 15 sub-fractions. Thin-layer chromatography (TLC) analysis of sub-fraction 15 revealed a single spot under UV light at 254 nm and 366 nm, and a positive spot after anisaldehyde staining. The melting point analysis indicated decomposition at 240°C, suggesting that sub-fraction 15 was pure. This sub-fraction was labeled as DS-HX-F1-SF15. The isolated compound was identified through the Wilstätter test, UV-Vis spectroscopy, and FTIR analysis, and was presumed to belong to the flavonoid class subclass flavanol. Its antibacterial activity was evaluated based on its Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). The MIC values were 100 ppm against B. cereus and S. aureus, and 125 ppm against
S. typhimurium and V. parahaemolyticus. The MBC values were >200 ppm for
