Isolasi Dan Uji Toksisitas Ekstrak Etil Asetat Dari Daun Bonai Antidesma Bunius (L.)
Antidesma bunius is a plant that belongs to the Euphorbiaceae family. In Indonesia, this plant is known as buni or bonai, which has properties as a traditional medicine. This study aims to isolate and characterize secondary metabolite compounds from ethyl acetate extract of A. bunius leaves, and to conduct toxicity tests using the brine shrimp lethality test (BSLT) method on the extract. Isolation was carried out using the vacuum liquid chromatography (VLC) method and continued with separation using flash column chromatography. The results obtained from this study were a white solid compound given the compound code AB-EA-2-F9. Purity analysis was carried out using a three-eluent system using thin layer chromatography (TLC) which obtained a single spot using a cerium sulfate spot detector. HPLC analysis was also carried out and peaks appeared at retention times of 2,5; 5,5 and there were 2 peaks at retention time of 19,00 minutes, this indicates that the compound is not pure. Characterization of the AB-EA-2-F9 compound was carried out using a UV-Vis spectrophotometer, FTIR and GC-MS. The UV-Vis spectrum of the AB-EA-2-F9 compound shows an absorption band at a wavelength of 204,94 with an absorbance value of 1,837. The IR spectrum of AB-EA-2-F9 compound shows the presence of functional groups at wave numbers 2918-2849 cm-1 (aliphatic C-H), 1706 cm-1 (carbonyl C=O), 1466 cm-1 (CH2), 1297 cm-1 (C-O) and 724 cm-1 (=C-H). The GC-MS spectrum of compound AB-EA-2-F9 showed a dominant peak at a retention time of 13,00 minutes and the compound name was detected as 1,11-Tridecadiene. Toxicity test was conducted on ethyl acetate extract of A.bunius. The toxicity test result obtained LC50 value of 3053,856 ppm, so it can be concluded that the extract is non-toxic.
