Isolasi Dan Uji Aktivitas Antijamur Senyawa Metabolit Sekunder Dari Ekstrak N-Heksana Tumbuhan Paku Sarang Burung (Asplenium Nidus L.)

Asplenium nidus, commonly known as the Bird’s Nest Fern, is a member of the Aspleniaceae family and has traditionally been used for medicinal purposes. Various communities have utilized this plant to alleviate fever, pain, and gastric ulcers. In addition to its therapeutic uses, A. nidus is also cultivated as an ornamental plant due to its distinctive leaf morphology and appealing coloration. This study aimed to isolate secondary metabolite compounds from the n-hexane extract of A. nidus and to evaluate their antifungal activity against pathogenic fungal Candida albicans, Candida glabrata, and Candida krusei, using the microdilution method. The research began with the maceration of A. nidus leaves in methanol. The resulting methanol extract was subsequently partitioned with n-hexane, dichloromethane, and ethyl acetate to separate compounds based on polarity. The n-hexane extract was further fractionated using flash column chromatography, yielding 23 major fractions based on thin layer chromatography (TLC) analysis. During this process, a yellow precipitate was observed in Fraction 20, which was subsequently recrystallized to yield a pure compound, designated as PSB-EH-F20. The purity of PSB-EH-F20 was confirmed by the presence of a single spot on the TLC plate after staining with anisaldehyde reagent, and a melting point range of 285-287°C. Spectroscopic identification using fourier transform infrared (FTIR) spectroscopy and comparison of Rf values with a standard compound indicated that PSB-EH-F20 was daucosterol. Antifungal testing revealed that PSB-EH-F20 inhibited the growth of all tested fungal species, with a minimum inhibitory concentration (MIC) of 100 ppm. The minimum fungicidal concentration (MFC) was found to be 200 ppm for C. albicans, while for C. glabrata and C. krusei, it exceeded 200 ppm. These findings suggest that PSB-EH-F20 possesses the ability to inhibit fungal growth at 100 ppm and exert fungicidal effects against C. albicans at 200 ppm. Therefore, PSB-EH-F20 holds potential as a natural antifungal agent for further development in pharmaceutical applications.
