Analisis Barkoding Dna Tumbuhan Pandan Asal Kubang, Kecamatan Siak Hulu, Kabupaten Kampar, Provinsi Riau Berdasarkan Sekuens Trnl-Trnl-Trnf Intergenic Spacer
ABSTRACT
Pandan plant from Kubang is a plant of the Pandanaceae family. Habitat and morphologically, pandan from Kubang has similar with Benstonea sp. from Pelalawan, but has a dissimilarity in the number of fruits. Pandan from Kubang has 2 fruits are located at the terminal bud, while Benstonea sp. from pelalawan has 1 fruit are located at the terminal of the stem. The identification of Pandanaceae group is difficult if only by observing morphology because it is difficult to find key characters from the Pandanaceae group of flowers and fruits in the field. So, it is necessary to analyze molecularly. This study aimed to analyze sequence of trnL-trnL-trnF Intergenic Spacer (IGS) in pandan plant from Kubang. DNA isolation, Polymerase Chain Reaction (PCR), electrophoresis, sequencing, and data analysis with the BioEdit, BLASTn, and MEGA 6 tools made up the research methodology. Research results showed that sequence of trnL-trnL-trnF IGS pandan from Kubang was an intron region and was 445 pb long. Benstonea sp. “Kajuik Lake” and Benstonea monticola has the most similarity (100%) to pandan plant from Kubang, according to the results of the BLASTn. Dendrogram analysis showed that pandan from Kubang are related to the Benstonea group. Pandan from Kubang is similar with Benstonea sp. “Kajuik Lake” based on geographic location and morphological similarity.
