Perkecambahan Dan Pertumbuhan Biji Anggrek Hutan Bunga Kuning (Coelogyne Rochussenii De Vriese) Secara In Vitro Dengan Penambahan Air Rebusan Kentang
Coelogyne rochussenii is a native Indonesian orchid species threatened with extinction due to widespread illegal harvesting from its natural habitat. Therefore, conservation efforts are essential, including propagation through in vitro culture. The Vacint and Went (VW) medium is a commonly used basic medium for orchid propagation in vitro. Still, it lacks vitamins, requiring the addition of organic substances such as potato decoction to enhance seed germination rates. This study aimed to investigate the seed germination response and protocorm growth of C. rochussenii orchids in vitro when treated with potato decoction, and to determine the optimal concentration of potato decoction for seed germination and protocorm growth. The study was conducted using a Completely Randomized Design (CRD) with five treatment levels (0, 50, 100, 150, 200 mL/L potato decoction and 150 mL/L coconut water in all VW medium treatments). The observed parameters included the protocorm emergence time, seedling growth response, and percentage of viable explants. Data were analyzed using Analysis of Variance (ANOVA), followed by Duncan’s Multiple Range Test (DMRT) at a 5% significance level. The results showed the seed germination phase of C. rochussenii consisted of five stages and a 100% explant viability. The addition of potato decoction at a concentration of 200 mL/L produced the best results, with the fastest protocorm emergence time of 25.2 days after planting (DAP), and the protocorm color was green (2.5 GY 7/6).
