Compounds from n-Hexane Root Extract of Tunjuk Langit (Helminthostachys
zeylanica) and Antifungal Activity Ugonin J and Ugonin K, supervised by Dr.
apt. Hilwan Yuda Teruna, M.Si and Dr. apt. Rudi Hendra Sy., M.Sc.
Helminthotachys zeylanica is a plant that belongs to a type of fern it this widespread
in rain forests in Indonesia such as Java, Sumatra and Borneo. This plant is used by
the community as a traditional medicine to cure dysentery, influenza and early stage
of tuberculosis. Several studies have successfully isolated secondary metabolite
compounds from H. zeylanica and have tested their bioactivities such as
antibacterial, antioxidant, antidiabetic and anticancer. The aim of this study was to
isolate the root extract of H. zeylanica and evaluate the antifungal bioavtivity of
ugonin J and ugonin K which have been isolated in previous studies in silico. The
n-hexane extract of H. zeylanica roots was separated using vacuum liquid
chromatography, flash chromatography and Sephadex® to obtain pure compounds.
Three pure compounds from this separation were tested for purity using thin layer
chromatography. The characterization of the extract was analyzed by spectroscopy
such as GC-MS and the characterization of the compounds was analyzed by FTIR
and H-NMR spectroscopy. Compounds C1-H-Hz were identified as unsaturated
fatty acids. For the antifungal activity test, ugonin J and ugonin K obtained in
previous studies showed that the compounds could potentially inhibit fungal
growth. Ugonin J and ugonin K were tested in silico with 14-α-demethylase
receptors (PDB ID: 5HS1, 1EA1 and 5TZ1) and 1,3-β-glucan (PDB ID: 4M8O).
Ugonin J and ugonin K indicated potential as antifungal drug candidates at 14-α-
demethylase and 1,3-β-glucan receptors and structural modifications were made for
better results.
