Isolasi Dan Karakterisasi Bakteri Pelarut Fosfat Dari Hutan Mangrove Bengkalis Sebagai Agen Biofertilizer

Coastal mangrove natural resources are very diverse, including microorganisms. Phosphate-solubilizing bacteria (PSB) are one of the microorganisms that can be utilized from the mangrove ecosystem. PSB can be used as a biofertilizer in agriculture to reduce excessive use of chemical fertilizers. The aim of this research is to isolate and characterize PSB from sediment and mangrove rhizosphere samples in Kelapapati Village, Bengkalis Regency, Riau. This research was conducted using the Pikovskaya medium, which has been adjusted to the conditions of the Bengkalis mangrove, including a pH of 6, a temperature of 30°C, and a salinity of 12.67 dS/m. Positive PSB results are characterized by the formation of a clear zone around the colony. Next, a phosphatase test and macroscopic characterization of PSB were conducted by observing morphology and microscopy through Gram staining. The highest PSE values were found in isolates SMB 1.2, SMB 2.2, and SMB 3.2 with an PSI of 2.8 from mangrove sediments, and isolate RA 1.3 from the rhizosphere of R. apiculata with an PSI of 2.7, which is categorized as moderate. The characterization results of all BPF isolates obtained from the Bengkalis mangrove show a diversity of morphological forms, but they have a similar cell shape, namely bacilli, with 17 isolates classified as gram-positive bacteria and 8 isolates classified as gram-negative. The success in obtaining PSB in this study indicates that the mangrove ecosystem in Kelapapati Village can be a source of PSB that has the potential to increase phosphate availability and has the ability to survive in changing conditions.
