Isolasi Senyawa Metabolit Sekunder Fraksi 6 Dari Ekstrak N-Heksana Daun Rukam (FlacourtiaRukam) Dan Uji Toksisitas Dengan Metode Brine Shrimp Lethality Test (Bslt)
Rukam is one of the species of Flacourtiaceae which is widely distributed in Sumatra and used as anti-diarrhea and anti-inflamatory ethnomedicine. Several species of this genus were reported to have antioxidant and cytotoxic activity. Phytochemical analysis showed that the leaf of rukam contained of flavonoids, saponins, fenolic, terpenoids and steroids. This study was aimed to isolate secondary metabolite from the leaf of rukam and to thest the toxicity of the compound by brrine shrimp lethality test (BSLT). A secondary metabolite (FR-H-
03) has been isolated from the leaf of F. rukam by flash chromatography. Isolation was done for fraction 6 of n-hexane extract to give 196 vials. Purification of vial numbers 112-113 obtained white crystaline compound (7 mg). The compound was characterized by FT-IR spectroscopy, which showed the presence of the OH group, aliphatic C-H, methylene group (CH2), methyl (CH3), cyclic alkene C=C and C-O (alcohol). The melting point was obtained at 260- 262oC. TLC analysis was done to compare FR-H-03 with daucosterol showed that FR-H-03 and daucosterol were similar. Toxicity assay showed that the n-hexane extract and 6 fraction were non-toxic with LC50 value 2529 ppm, 1527 ppm respectively, and FR-H-03 compound showed toxic activity with LC50 value 13 ppm.
