Isolasi Dan Uji Antibakteri Ekstrak Etil Asetat Dari Daun Bonai Antidesma Bunius (L.)
Antidesma bunius is one of the species belonging to the Euphorbiaceae family. Three species of Antidesma, A. madagascariensis, A. ghaesembilla and A. venosum have been widely reported to have antibacterial activity. This study aims to isolate and characterize secondary metabolite compounds, and identify the antibacterial activity of ethyl acetate extract of A. bunius bonai leaves using the disc diffusion method. Isolation was carried out using the vacuum liquid chromatography (VLC) method, flash column chromatography, followed by purification and characterization. The results of the study obtained a white solid compound coded AB-EA-2-F33. Purity analysis was carried out by thin layer chromatography using a three eluent system, melting point and high performance liquid chromatography (HPLC). Characterization of the AB-EA-2-F33 compound was carried out using a UV-Vis spectrophotometer, FTIR and GC-MS. The results of the UV-Vis analysis of the compound showed an absorption band at a wavelength of 205 nm with an absorbance value of 1.893. The IR spectrum shows the presence of functional groups at wave numbers 3268 cm-1 (O-H), 2955- 2850 cm-1 (aliphatic C-H), 1735 cm-1 (C=C), 1652 cm-1 (C=C) and 1240 cm-1 (C-O). The results of GC-MS analysis of AB-EA-2-F33 compound are classified as fatty acids containing hexadecanoic acid and trans-13-octadecanoic acid compounds. The results of antibacterial activity tests using the disc diffusion method showed that this extract is resistant to Staphylococcus aureus and Escherichia coli bacteria with a concentration of 1000 ppm.
