Respon Pertumbuhan Eksplan Jeruk Siam (Citrus Nobilis L.) Pada Media Ms Dengan Penambahan Bap Dan Naa Secara In Vitro
ABSTRACT
The cultivation of Siamese orange (Citrus nobilis L.) in Kampar Regency has the main obstacle, namely the attack of diplodia disease caused by Botryodiplodia theobromae causing the plants to die so that citrus production decreases. Conventional plant propagation generally takes a long time and often experiences technical problems in the field. In vitro culture techniques on aseptic media with the addition of growth regulators are an option in an effort to provide seeds of a plant. This study aims to determine the effect of the interaction of BAP and NAA on MS media on the growth of Siamese orange explants. The research was conducted at the Laboratory of Plant Biotechnology, Faculty of Agriculture, Riau University from November 2021 to February 2022. The experiment used a randomized block design with two factors, namely four levels of BAP 0, 1, 3, and 5 ppm and four levels of NAA namely 0, 0.1, 0.2, and 0.3 ppm with three replications. The results showed that the treatment of 0.1 ppm NAA without BAP gave the best response at the time of emergence of roots 17 DAP, treatment of 0.2 ppm NAA without BAP gave the best response at the number of roots 5.58 strands, treatment 1 ppm BAP and 0.3 ppm NAA gave the best response to an increase in shoot height 0.92 cm, and the treatment of 1 ppm BAP and 0.1 ppm NAA gave the best response to an increase in the number of leaves 2.69 strands.
