Seleksi Dan Uji Daya Antagonis Jamur Endofit Asal Tanaman Mahoni (Swietenia Macrophylla King.) Terhadap Jamur Rigidoporus Microporus (Swartz:Fr) Van Ov. Penyebab Penyakit Jamur Akar Putih Pada Tanaman Karet
ABSTRACT
Endophytic fungi has been much reported to be applied to control fungal disesases on plants. The fungi can be isolated from almost of the plants tissues, included mahogany plant and then can be applied to control fungal diseases on other plants, such as white root disease on rubber plant. The research was conducted to screen, evaluate the antagonistic activity of endophytic fungi isolated from mahogany plant and obtain isolates that have high antagonistic activity against R. microporus and identify them to the genus level. The research was conducted at the Plant Disease Laboratory, Faculty of Agriculture, Riau University. The study used some methods : exploration (isolation and purification of the endophytic fungi of mahogany plants and pathogenic fungi, R. microporus), observation (hypovirulence test, hyperparasitism test, and identification of th endophytic fungal isolates), and experiment (antagonistic activity test and endophytic fungal growth test). The data from macroscopic characteristics, hypovirulence test, hyperparasitic type and identification of the endophytic fungi of mahogany plants were analyzed descriptively. The data from antagonistic activity, diameter and growth rate of the endophytic fungi were analyzed with analyses of variance using SPSS application. The results of the endophytic fungi isolation from mahogany plants obtained 12 isolates, five isolates from roots (M1, M3, M5, M9 and M10), four isolates from stems (M4, M7, M8 and M11) and three isolates from leaves (M2, M6 and M12). Nine isolates were hypovirulent (M1, M2, M3, M4, M5, M7, M9, M10 and M12) and three isolates (M6, M8 and M11) hypervirulent fungi. Isolates M2, M5, M12, M7 and M9 were able to inhibitR. microporus growth with the rate of 68.84%, 67.01%, 64.07%, 62.47% and 58.60%, respectively. The largest colony diameter and highest growth rate were isolate M7 (90.00 mm and 36.31 mm.day-1). The hyperparasitic types of the endophytic fungi that have high antagonistic activity against R. microporus were entanglement (M2, M9 and M12) and attachment (M5 and M7). The identification results of the endophytic fungi showed that isolates M2, M9 and M12 belonged to the genus Trichoderma, isolates M5 and M7 belonged to the genus Aspergillus.
