Untargeted Metabolomics Dan Uji Aktivitas Antibakteri Dari Tanaman Tunjuk Langit (Helminthostachys Zeylanica)
Tunjuk Langit (H. zeylanica) is a plant that has been used empirically as a
traditional medicine by the community. From several studies they were reported
to produce several secondary metabolites with various pharmacological effects.
Metabolomics is a technology that is currently being developed to investigate the
relationship between plant metabolism and pharmacological effects. Untargeted
metabolomics combined with chemometrics can provide a comprehensive
overview of a plant's metabolites and predict the compounds that play a role in
their pharmacological activities. The purpose of this study is to identify secondary
metabolites and test their antibacterial activities in H. zeylanica roots, stems, and
fronds. The Kirby-Bauer method was used to test the antibacterial activity of
methanol, n-hexane, dichloromethane, ethyl acetate, and water extracts. The
broth and agar dilution method was used to determine the Minimum Inhibition
Concentration (MIC) and Minimum Bactericidal Concentration (MBC).
MetaboAnalyst 5.0 was used for chemometric analysis and UHPLC-Q-Orbitrap
HRMS was applied for untargeted metabolomics.The antibacterial test results
showed that the dichloromethane extract could inhibit the growth of some
bacteria. Test against Listeria innocua ATCC 33090, Micrococcus luteus ATCC
10240, Salmonella typhimurium ATCC 14028, Vibrio parahaemolyticus ATCC
17802, Vibrio cholerae inaba, Escherichia coli ATCC 25922, Shigella sonei
ATCC 9290 (for roots and stems), and Bordetella bronchiseptica ATCC 4617
showed MIC values of 31.25-500 μg/mL and MBC values of 62.5-500 μg/mL.
According to the results of the metabolomics analysis, the compound acting as an
antibacterial in the roots is NCGC00385829-01!2-(3,4-dihydroxyphenyl)-6-[(2E)-
3,7-dimethylocta-2,6-dienyl]-5,7-dihydroxy-2,3-dihydrochromen-4-one, the
compound in the stems is oleyl sarcosin, and the compound in the fronds is
loliolide dan [5-(2-methylimidazo[1,2-a]pyridin-3-yl)-1H-pyrazol-1-yl][4-
(trifluoromethyl) phenyl]methanone, as well as one unknown compound.
