Aktivitas Antibakteri Dan Antioksidan Dari Metabolit Sekunder Yang Diisolasi Dari Tumbuhan Paku Pyrosia Longifolia

Pyrrosia is a genus of tropical forest-dwelling epiphytic plants. This plant is
widely used in traditional medicine due to its antioxidant, anti-diabetic, antiinflammatory,
and antibacterial qualities. However, its antibacterial and
antioxidant properties have never been documented. This research aims to assess
the antibacterial and antioxidant properties of P. longifolia extract and to isolate
secondary metabolites from the active extract. For the purposes of this study, the
compounds were isolated using the bioassay-guided technique. In addition,
antibacterial activity was determined using diffusion and microdilution techniques
against several pathogenic bacteria, and antioxidant activity was determined using
free radical scavenging against the DPPH radical. Utilize chromatography
methods including vacuum liquid and flash chromatography to obtain pure
compounds. Six isolated compounds and their purity of the active fraction were
analyzed using melting point, thin layer chromatography, and HPLC techniques.
Spectroscopy analyses such as UV-Vis, FTIR, and NMR were employed to
elucidate the chemical structures of isolated compounds. The compounds were
identified as naringin (30), catechin (31), quercetin (32), rutin (33), kaempferol
(34), and mangiferin (35) based on the analysis of spectroscopy data. To
determine the absolute configuration of naringin (30) and catechin (31), a circular
dichroism spectrometer was applied to the compounds. The results showed that
the compounds (2S) naringin (30) and (2R, 3S) catechin (31) were identified. All
of the compounds were able to inhibit bacterial growth with MIC values ranging
from 7,8 μg/mL to 250 μg/mL and MBC values ranging from 15,6 μg/mL to 250
μg/mL. Moreover, all the compounds exhibited antioxidant activity with IC50
values ranging from 14,61 to 36,11 ppm. These findings suggest that P. longifolia
could be utilized as an antibacterial and antioxidant agent.
