Isolasi Dan Uji Antibakteri Senyawa Flavonoid Dari Ekstrak Metanol Jamur Endofit Botryosphaeria Rhodina
SUMMARY
Endophytic fungi are microorganisms that reside in plant tissues, have no negative effects on their hosts, and can produce large quantities of secondary metabolites identical to those of their hosts. The endophytic fungus B. rhodina is a member of the genus Botryosphaeria, which can grow in a variety of climates worldwide. Multiple species of this genus have been reported to possess antimicrobial properties. The purpose of this investigation was to isolate secondary metabolites from the methanol extract of the endophytic fungus B. rhodina and assess their antibacterial activity. The endophytic fungus was fermented on rice solid media for 9 days, followed by maceration in ethyl acetate solvent to obtain secondary metabolites. The crude macerate of ethyl acetate was then extracted using n-hexane and methanol. The methanol extract was separated by Vacuum Liquid Chromatography (VLC) into seven impure major fractions (FI-FVII). Using Flash Chromatography, FVI was purified and four principal sub fractions were obtained (SF1-SF4). The TLC chromatogram for sub fraction 3 (SF3) yielded one spot visible with a 366 nm UV lamp, the melting point test yielded a value between 210-212°C and HPLC results obtained one peak at a retention time of 13.85 minutes, this indicates that the compound was pure SF3 with the code 21-07-EM-FVI-SF3-60/170. The obtained compound was characterized via UV-Vis and FTIR. There are two absorption bands at a wavelength of 281 nm and 366 nm with absorbance values of 0.150 and 0.087. The FTIR analysis of the compound show absorption at wave numbers 3260 cm-1, 2852 cm-1, 1654 cm-1, 1565 cm-1 and 1077 cm-1 revealed the presence of O-H, C-H, C=O, C=C and alcohol C-O groups. The antibacterial activity of Gram-positive and Gram-negative bacteria was examined. The compound 21-07-EM-FVI-SF3-60/170 has antibacterial activity against S. aureus ATCC 6538, B. subtilis ATCC 6633, L. monocytogenes ATCC 7644, V. alginolyticus ATCC 14028 and S. Typhimurium ATCC 17749 with MIC values of 25 μg/mL, 50 μg/mL, 25 μg/mL, 25 μg/mL, 100 μg/mL respectively with MBC values of 100 μg/mL, 100 μg/mL, 100 μg/mL, 50 μg/mL and > 100 μg/mL.
