Isolasi Dan Uji Toksisitas Senyawa Metabolit Sekunder Dari Fraksi Etil Asetat Kulit Batang Tumbuhan Pulai Alstonia SCHOLARIS

SUMMARY
Alstonia scholaris is known in Indonesia as pulai. This plant belongs to the Apocynaceae family and it is widespread in the tropical regions of Africa and Asia. Alstonia scholaris (L.) R.Br., commonly called blackboard tree, devil tree in South and Southeast Asia.This plant is widely used in traditional medicine, especially in India and Southeast Asia, including India. Various Indian traditional medicines such as Ayurveda, Unani, Homeopathy, and Sidha papers that A. scholaris was used to treat asthma, malaria, fever, dysentery, diarrhea, epilepsy, skin diseases, and snake bites. This study aims to isolate and characterize pure compounds obtained from the ethyl acetate fraction and to test their toxicity using the BSLT (brine shrimp lethality test) method. Isolation was carried out by maceration and followed by partitioning. Separation of isolated compounds was carried out by flash column chromatography which produced 12 fractions, crystals were produced in fraction 5 and coded ASc_E_34. Further separation was carried out on fraction 4 by further Sephadex LH-20 column chromatography and produced crystals coded ASc_Sph_3-11. The crystals have melting points of 128-130 °C and 136-140 °C. The UV-Vis spectrum of the compound ASc_E_34 (λ) 285 nm (A) 0.116. The IR spectra have absorption bands at 3587; 3566 cm-1 (N-H), 2941; 2849 cm-1 (aliphatic C-H), 1683-1512 cm-1 (C=C) and 1272; 1143 cm-1(C-O alcohol) which is reinforced by the presence of a widening absorption band in the 3000-3500 cm-1 region which indicates OH, 847 cm-1 (C-H). UV-Vis spectrum of the compound ASc_Sph_3-11 (λ) 288 nm (A) 0.280; (λ) 326 nm (A) 0.294. The IR spectra have absorption bands at 2935; 2844 cm-1 (aliphatic C-H), 1595 cm-1 (C=C),1272-1013 cm-1(C-O alcohol). Based on the results of the analysis, the two compounds was alkaloid compounds. The toxicity test stated that both compounds were not toxic by showing an LC50 value > 1000 ppm.
