Isolasi Dan Uji Toksisitas Senyawa Metabolit Sekunder Dari Ekstrak N-Heksana Kulit Batang Kelor Moringa Oleifera
Moringa oleifera plant is a plant that is widely used as a medicinal plant that comes from the Moringa genus and belongs to the Moringaceae family. This study aims to isolate the secondary metabolite compound n-hexane extract from the stem bark of M. oleifera moringa and analyze its toxicity activity using the brine shrimp lethality test (BSLT) method. This study aims to isolate the secondary metabolites of n-hexane extract from the stem bark of M. oleifera and analyze their toxicity using the brine shrimp lethality test (BSLT) method. Fractions 2 and 3 were combined and then separated by flash chromatography to obtain 112 vials of separation. Vials 22, 28 and 30 were in the form of crystals with some impuritis, then combined and separated again by flash chromatography and 51 vials were obtained. Crystals formed in vials 4 and vials 5 which were then re-crystallized. From the re-crystallization process it was obtained two white crystalline solids with different physical forms. First one was coded MO-HF1-V4 with a physical form of plates (20.4 mg) and seconed one was coded MO-HF1-V5 with a physical form of powder (3.4 mg). The purity of the crystals was analyzed using TLC which. It showed that the two compounds were similar and the melting point test obtained from MO-HF1-V4 was 120-122oC (not calibrated, so it can be different) while MO-HF1-V5 was obtained 128-130oC (not calibrated, so it can be different). The FTIR spectrum showeds that there were some bands at 3301 cm-1 (O-H), 2957-2851 cm-1 (C-H), 1465 cm-1 (CH2), 1379 cm-1 (CH3) and 1049 cm-1
(C-O). The results of the data obtained from the FTIR spectroscopy and the TLC
comparison test with the β-sitosterol compound show that these two crystals have similarities to the β-sitosterol compound. Toxicity analysis of n-hexane extract using the BSLT method showed that the n-hexane extract of M. oleifera stem bark was toxic, with an LC50 value of 321.66 ppm.
