Isolasi, Uji Antioksidan Dan Antidiabetes Senyawa Metabolit Sekunder Tumbuhan Paku Tanduk Rusa (Platycerium Bifurcatum)
SUMMARY
Platycerium bifurcatum is an epiphytic plant that belongs to the Polypodiaceae family. Other species such as P. superbum, P. stemaria have been reported as potential sources of antioxidants, antibacterial, anti-inflammatory and anticancer, but information and research on the potential bioactivity of P. bifurcatum was limited. This study aims to isolated secondary metabolite compounds and determine the antioxidant and antidiabetic activities of ethyl acetate extract of P.bifurcatum. Secondary metabolites were obtained from extraction by maceration method used methanol solvent. The methanol crude macerate was followed by liquid-liquid extraction used n-hexane, dichloromethane (DCM), and ethyl acetate. The ethyl acetate extract was separated used VLC . The VLC results, there was a yellow precipitate in fraction 4 and then recrystallization was carried out, so that fraction 4 obtained one stain which was seen at UV lamp 366 nm and 264 nm, and melting point test used Fisher-Jhons tool obtained melting point at temperature 272-274 ℃, and coded 22-09-C-FIV. Fraction 6 was purified by flash chromatography column. Sub-fraction 45 contained a yellow precipitate and then recrystallized, so that sub-fraction 45 obtained one stain which was seen at UV lamp 366 nm and 264 nm, and melting point test used Fisher-Jhons tool obtained melting point at temperature 258-260 ℃ and coded 22-09-C-FVI-SF 45/215. Pure compounds were characterized used UV-Vis, FTIR, and NMR. UV-Vis results showed that compound 22-09-C-FIV had absorption peaks at a wavelength of 205 nm, 266 nm, 364,5 nm with absorbance of 0,624 ; 0,4 ; 0,473 and for compound 09-C-FVI-SF 45/215 at a wavelength of 206 nm, 255 nm, 360 nm with absorbance of 0,511; 0,259; 0,277. The FTIR characterization results of both compounds showed the presence of O-H, C-H aromatic, C=O, C=C, and C-O groups. The characterization results show that both compounds are flavonoids, a class of polyphenols in which compound 22-09-C-FIV was kaemferol and 09-C-FVI-SF 45/215 was apigenin. Antioxidant assay used DPPH method with IC50 values for kaemferol (21 μg/ml) and apigenin (14 μg/ml). Antidiabetic assay was used the α-glucosidase enzyme inhibition method with IC50 values on kaemferol (56.6367 μg/ml) and apigenin (>250 μl).
