Isolasi Dan Aktivitas Inhibisi Enzim Α-Glukosidase Oleh Senyawa Metabolit Sekunder Dari Ekstrak Etil AsetatTumbuhan Paku (Stenochlaena Palustris)
SUMMARY
Stenochlaena palustris, is one of a fern that can bu use as antioxidant,
antimicrobial, antianemia, and antidiabetic agent. S. palustris may be a promising
anti-diabetic drug, but little is known about its secondary metabolites. This
research isolated S. palustris ethyl acetate extract by maceration method with
methanol was followed by partition with n-hexane, dichloromethane, and ethyl
acetate. The ethyl acetate extract of S. palustris was then purified using the
vacuum liquid chromatography (VLC) method to produce 16 fractions (F1-F16),
which were then combined using thin layer chromatography (TLC) by Rf value.
Furthoremore, fraction F5 was purified by using gel chromatogprahy with
Sephadex-LH 20 as stationary phase and yielded 50 subfractions (SF1-SF10).
Subfractions SF7 and SF9 produced pure compounds, which were coded 19-04-CFV-
SF7 and 19-04-C-FV-SF9, respectively, the obtained compounds were then
characterized using UV-Vis, FTIR, and NMR. The results of the UV-Vis
characterization of compound 19-04-C-FV-SF7 showed several maximum
absorption band at wavelengths of 267 nm, 365 nm with absorbances of 0.268,
0.273 respectively and for compound 19-04-C-FV-SF9 showed maximum
absorption band at wavelengths of 368 nm with absorbances of 0.166, both result
indicating the presence of conjugated double bonds. The results of the FTIR
analysis of these two compounds showed the presence of O-H groups, C-H alkane
groups, C=C groups, and C-O groups of aromatic esters. Compound 19-04-C-FVSF9
it is indicated to be a kaemperol compound, while compound 19-04-C-FVSF7
is identified as quercetin based on published data. The compounds were
tested for α-glucosidase inhibition activity, and the IC50 for compound 19-04-CFV-
SF7 was 80.80 μg/mL and 56.64 μg/mL for compound 19-04-C-FV-SF9.
